DNA Test Requests, Results & Feedback

DNA Test Requests 

When submitting a DNA test request, consideration should be given to the choice of laboratory, sample type and the intended outcome of the test. DNA test requests can be submitted online through Helical or via email to [email protected] 

Once a submission has been reviewed and approved by a Member Services Officer (MSO), an automated email is sent to the email address recorded on the membership. This email includes an Acknowledgement Letter, which outlines the details of the submitted request as well as the laboratory postage information. 

The Acknowledgement Letter must be printed and submitted with the samples which are sent directly to the nominated laboratory.

Results & Feedback 

Once samples are received by the laboratory, DNA testing typically takes approximately 6–8 weeks for results to be returned to the Australian Wagyu Association (AWA). In most cases, genomic SNP results used for parent verification are received earlier than genetic condition results. 

Upon receipt, genomic SNP results are used by the AWA for in-house parent verification, ensuring a controlled and consistent process. Once parent verification is complete, an email is sent to the membership email address. This includes parentage results, a pre-filled registration form, and any Parentage Awaiting Feedback items requiring further action. 

Both parentage and genetic condition results can also be viewed directly in Helical, where the progress bar displayed alongside each test request indicates how many results have been returned. 

Genetic Conditions Results 

Animals that have been tested will have definitive results being Free, Carrier or Affected. Animals that haven’t been tested may show percentage probabilities based on GeneProb or show as Free Untested (FU) based on Pedigree information.

Parentage Awaiting Feedback Report 

Where clarification is required, a Parentage Awaiting Feedback report is issued alongside the results via email. This information is also accessible within Helical, allowing members to review and address outstanding items at any time. 

To resolve DNA feedback items, members must email [email protected] with the relevant animal details and proposed resolution. Common issues identified in Parentage Awaiting Feedback reports include: 

  • Nominated parents not verifying or concerns regarding parent suitability 
  • Mismatch between the genotyped sex and the sex recorded at the time of test request 

The purpose of this feedback process is to ensure the highest possible level of accuracy in parentage determination and maintain the integrity of herd records. 

Call Rates 

The call rate refers to the integrity of the genotype and overall quality of the DNA sample. A low call rate may affect: 

  • Parentage analysis involving the animal and its progeny; and 
  • The ability for the animal’s genotype to be included in the genetic evaluation and receive genomically enhanced WBVs. 

Call rates provide an indication of genotype quality and can help identify potential issues with the DNA sample or testing outcome.

Recollects and Failed Samples 

A sample may be flagged as a recollect if it arrives at the laboratory and is visually assessed as unsuitable for testing before analysis begins. Typical reasons include: 

  • TSU or hair card collector is empty 
  • TSU has leaked and the tissue sample is shriveled up and starting to degrade 
  • No hair follicles are present on the hair sample 
  • Insufficient hair follicles have been collected (a minimum of 40 follicles is recommended to allow for potential retesting) 
  • Hair sample collected before animal is three months of age and the follicles are not developed 
  • Hair sample is heavily soiled 
  • Hair sample is wet and/or mouldy 

These samples do not proceed to laboratory testing and are immediately flagged as recollects. 

failed sample differs from a recollect. Failed samples have passed the initial visual assessment and proceeded through laboratory testing. However, despite completing the testing process, the sample does not produce results of sufficient quality to generate conclusive outcomes. In these cases, the sample is classified as failed and a new sample must be collected and submitted for testing. 

Common Reasons why a sample might fail: 

  • Biological contamination – for example, heavily soiled samples contaminated with manure, dirt, or other organic material. 
  • Chemical contamination – for example, exposure to insect repellents, disinfectants, cleaning agents, or other chemicals. 
  • Improper storage – for example, prolonged exposure to heat, such as leaving samples in a vehicle or direct sunlight for extended periods.

DNA parentage analysis within the Australian Wagyu Association (AWA) is conducted in two stages: parent verification and, where required, parent discovery.
DNA testing is a mandatory requirement for registering animals and accessing Wagyu Breeding Values (WBVs) within the database.
The Australian Wagyu Association (AWA) accepts the following sample types: -Tissue Sampling Unit (TSU) -Tail hair samples -Semen samples
When extra samples are received by the lab, the lab will store (c-bank) these samples. Members can now submit a DNA test request for any animals that have a stored sample recorded. Follow the steps below to complete your submission to test a stored sample.

It is a condition of use of this database that you accept these terms and conditions. 

It is important that you appreciate when accessing the Australian Wagyu Association Limited (AWA) database (which may be hosted on an external website) that the information contained on the AWA database, including but not limited to pedigree, DNA information, Wagyu Breeding Values (WBVs) and Index values (together called Database Information), is based on data supplied by AWA members and/or third parties. 

Whilst sincere effort is made to ensure the Database Information is accurate and complete, to the extent permitted by law, AWA, its officers and employees, shall not be responsible for its use or interpretation, and you use the Database Information at your own risk. Please be aware of the following limitations of the Database Information.  

Regarding WBVs and Index values, it is important to appreciate, and you need to be aware that: 

  • WBVs are derived using genetic evaluation technology developed for the AWA, using information contained within the AWA database and parameters derived from it.
  • WBVs are estimates of genetic potential of individual animals and may change with addition of new data or with new parameters.
  • WBVs are genetic merit estimates only and may not reflect the raw animal phenotype.
  • WBVs can only be directly compared to other WBVs calculated in the same AWA genetic analysis.

Regarding pedigree and DNA testing results submitted to the AWA, it is important to appreciate, and you need to be aware that: 

  • Animals whose registration identifiers start with PED are Pedigree Recorded Animals. What this means is that the details which AWA used to register the animal were obtained from a Recognised Wagyu Registry Organisation – in other words a Wagyu breed association based outside of Australia. AWA has not independently verified the information with respect to the animal or the details recorded about the animal. You should read AWA’s by-laws which set out details regarding the definition of a Pedigree Recorded Animal. 
  • Pedigree and DNA data submitted and supplied to AWA may have errors in it which cannot be detected without further DNA testing. 
  • Technology may have advanced since a particular test was undertaken so that genetic issues or inaccuracies which were previously not detectable are now able to be detected by current testing technology (i.e. if the animal was tested again with current technology). 
  • AWA estimates that less than 1% of the pedigree entries, ownership or breeding details in the AWA Herdbook may have errors or which may be misleading. For this reason, users ought to consider if they need to obtain independent testing of the relevant animal (if possible) to ensure that the data is accurate. 

Regarding prefectural content, it is important to appreciate, and you need to be aware that: 

  • Prefectural content is based on the estimation of prefectural origin from Japanese breeding records of 201 foundation sires and 168 foundation dams. As genotype-based parent verification is not used in Japan, and full Japanese registration certificates are not available for all foundation animals, exact prefectural composition for these sires and dams cannot be validated. 
  • The calculation of prefectural content for Australian Herdbook animals relies on the accuracy of pedigree records and DNA samples provided by AWA members.   

Regarding DNA testing and genetic condition results submitted to the AWA, it is important to appreciate, and you need to be aware that: 

  • As genetic testing results are provided by independent DNA testing laboratories at the request of AWA members, AWA relies solely on the accuracy of the reporting of genetic testing as completed by the laboratories. 
  • AWA uses GeneProb software to calculate the likelihood of pedigree animals being a carrier of genetic conditions.  This likelihood is an estimate based on the available genetic testing results. 

If you consider that you do not understand or appreciate the nature and/or implications of the data provided on this website or the WBVs of a particular animal, then AWA strongly recommends that you seek expert advice.  Note there is substantial technical information and articles on the Wagyu website to assist our members.
